Submitting Your Samples

Customers have the following options for submitting samples for DNA sequencing:

1.) Perform the sequencing reaction and clean-up in your lab. This means that you purchase a BigDye terminator kit from our facility and run the cycle sequencing reaction and post clean-up. We recommend using Princeton Separations Centri-Sep spin columns or Abgene Dye Terminator Removal Plates. Drop off your sample in the form of a dried pellet in 1.5 ml eppendorf tube along with your request form. Your dnalims request order will have Seq_Drop_Off selected under Service Requested, and have no selected under Spin-Column Clean-up.


2.) Perform the sequencing reaction in your lab and request clean-up by our facility. This means that you purchase a BigDye terminator kit from our facility and run the cycle sequencing reaction without performing post clean-up. Drop off your 20ul sample in a PCR tube along with your request form. Our facility will perform the clean-up using an Agencourt CleanSeq Magnetic bead kit. Your dnalims request order will have Seq_Drop_Off selected under Service Requested, and have yes selected under Spin-Column Clean-up.


3.) Request that our facility perform the sequencing reaction (using standard primers) and post clean-up. This means that you bring the appropriate concentration of template (20-40 ng PCR product for every 200 base pairs or 600-1000ng plasmid) in a 22ul volume in a 1.5 ml eppenforf tube along with your request form. We will add the standard primer you have selected (M13F, M13R, T7, T3, SP6, KS, SK) on your dnaLIMS request form. Our facility will add the BigDye terminator and perform the cycle sequencing reaction and post clean-up using an Agencourt CleanSeq Magnetic bead kit. Your dnalims request order will have Standard selected under Service Requested.


4.) Request that our facility perform the sequencing reaction (using your primer) and post clean-up. This means that you bring the appropriate concentration of template (20-40 ng PCR product for every 200 base pairs or 600-1000 ng plasmid) and primer (8 pmol) pre-mixed in a 24ul volume in a 1.5 ml. eppendorf tube along with your request form. Our facility will add the BigDye terminator only and perform the cycle sequencing reaction and post cleanup using the Agencourt CleanSeq Magnetic bead kit. Your dnalims request order will have Pre-Mixed selected under Service Requested.
